At the time of the 2010 Census, Wilmot had a population of 492 people. This makes Wilmot the county's 2nd most populous community.

The population of Wilmot represents 0.06% of the total population of South Dakota (which has 814,180 people), making it the state's 154th most populous community.

With a 2010 count of 492 people, the population of Wilmot decreased 9.39% from the 2000 Census (which had a count of 543 people).
