Taken from the 2019 Census Estimates, Burke had an population of 585 people. This is a decrease of 3.15% since the 2010 Census (or a decrease of 13.46% since the 2000 Census).

At the time of the 2010 Census, Burke had a population of 604 people (or 14.14% of the total population of Gregory County). This makes Burke the county's 2nd most populous community.

The population of Burke represents 0.07% of the population of South Dakota (which has 814,180 people), making it the state's 134th most populous community.

With a 2010 count of 604 people, the population of Burke decreased 10.65% from the 2000 Census (with a count of 676 people).
