At the time of the 2010 Census, Buffalo had a population of 330 people (or 26.29% of the total population of Harding County). This makes Buffalo the county's 2nd most populous community.

The population of Buffalo represents 0.04% of the population of South Dakota (which has 814,180 people), making it the state's 200th most populous community.

With a 2010 count of 330 people, the population of Buffalo decreased 13.16% from the 2000 Census (which had a count of 380 people).

From our notes, the earliest published mention we've found for Buffalo was in the document titled List of Post Offices from Cameron Blevins and Richard Helbock.<7>

From that list, the Buffalo post office opened in 1909.
