At the time of the 2010 Census, Kingstree had a population of 3,328 people (or 9.67% of the total population of Williamsburg County). This makes Kingstree the county's most populous community.

The population of Kingstree represents 0.07% of the total population of South Carolina (which has 4,625,364 people), making it the state's 82nd most populous community.

With a 2010 count of 3,328 people, the population of Kingstree decreased 4.81% from the 2000 Census (which had a count of 3,496 people).
