Greene County was founded in 1833. Greene was created by taking area from Crawford and Wayne Counties.

From the Census Estimates for 2019, <1> Greene had a population of 293,499 people. This is an increase of 6.66% since the last Census (which gave a population of 275,174 people in 2010).

At the time of the 2010 Census, Greene represented 4.59% of the total population of Missouri.
